Nuclear Magnetic Resonance (NMR)

  • NMR spectroscopy is one of the most important instruments to identify the molecular structure, composition and dynamics of organic compounds. The same is analyzed by observing and measuring the interaction of nuclear spins when placed in a superconducting magnetic field. DSK’s in-house NMR Spectrometer can perform various experiments as per the following details.
  • NMR Spectrometer : 400 MHz Jeol Make with 24 sample Autosampler and is equipped with a Multiprobe.
  • The NMR experiments can be performed in 1D, 2D & 3D depending on the complexity of the system being studied.
  • 1D experiments such as1H, 13C, 19F, 31P, 15N, 39K, 109Ag can be run with this NMR instrument.
  • 2D experiments such as COSY, NOESY, HSQC, HMBC, TOCSY can also be performed with this instrument.

Mass Directed Auto Purification (MDAP) System

  • MDAP is a combination of both Prep HPLC along with LC-MS system.
  • Mass-directed preparative chromatography provides higher sensitivity and more specificity than traditional UV-based detection.
  • Mass detection can easily be 100 times more sensitive than UV and in Scan Mode, the mass detector delivers unambiguous molecular information about every peak.
  • We can program peak collection based the molecular weight, or the mass to charge ratio – to collect the correct peak of interest.
  • DSK’single quadrupole Shimadzu LCMS-2020 mass spectrometer along with the Nexera Prep HPLC system utilizes the patented ultrafast (UF) technology, delivers more information about the preparative runs and enables us to collect the right peak.
  • High yield of fraction collection is achieved by sending only a small amount of sample into the highly sensitive LC/MS using automated proportioning valve.
  • UF switching, UF scanning and High sensitivity of LCMS-2020 make it an ideal choice for an efficient and reliable fraction of even complex mixtures.
  • The capacity of purification ranges from 10mg to 2g.

Liquid chromatography - Mass Spectrometry (LC-MS)

  • LC / MS is an analytical technique that combines the separation power of HPLC with the direct mass measurement of a mass spectrometer as the detector.
  • HPLC can separate a wide range of compounds, while the mass detector provides valuable information about the molecular weight, structure, identity, quantity, and purity of a given compound.
  • DSK’ Shimadzu LCMS-2020 Mass Spectrometer along with the Nexera Prep HPLC system can detect compounds in the mass ranges of 50-2000m/z. It has a dual ion source which can continuously perform two types of ionization, ESI and APCI simultaneously.
  • DSK’ Waters HPLC Alliance with QDaMass Detector can detect compounds in the mass rangesof 50-1250m/z with only ESI as the single ionization source.

High-Performance Liquid Chromatography (HPLC)

  • HPLC instrument is an analytical chemistry technique applied to separate, identify, and quantify semi- and non-volatile compounds in liquid samples. It is used for the characterization, purification, and quantitation of compounds.
  • HPLC are used to analyze compounds with chromophore functional groups. The related substance, assay & enantiomeric purity can be studied.
  • DSK has a few Waters HPLC Alliance with PDA detectors. The operating system pressure is up to 4000psi.
  • DSK has many Agilent HPLC 1260 Infinity-II series with PDA detector. The operating system pressure is up to 600/800bar.

HPLC with ELSD & RI Detectors

  • HPLC with Evaporative Light Scattering Detectors (ELSD) are aerosol-based detectors which are used to analyze compounds with non-Chromophore functional groups.
  • DSK uses an Agilent 1260 Infinity II with PDA & ELSD for such compounds.
  • HPLC with RI Detector and Gel Permeation Chromatography (GPC) Software are used for those compounds which cannot be deducted by UV detectors such as polymers. The molecular weight ranging from 160 to 500000 g/mol can be identified in these systems.
  • DSK uses an Agilent 1260 Infinity II with PDA, RI Detector & GPC Software for such polymer compounds.

Gas Chromatography (GC)

  • Gas chromatography (GC) is an analytical technique for separating and analyzing compounds that can be vaporized without decomposition. Typical uses of GC include testing the purity of a particular substance or separating the different components of a mixture.
  • DSK uses Agilent 8890 Gas Chromatography (GC) with Flame Ionization Detector (FID) to test those compounds with anon-Chromophore functional group and lowerboiling points.
  • DSK uses an Agilent 7890B GC with a 7697A Headspace Sampler system to check the Residual Solvents in organic compounds in ppm levels.

Polarimeter

  • Polarimeter is an instrument used to determine the Optical Rotation and Specific Optical Rotation (SOR) of a compound. The SOR helps in identifying, characterizing and determining the purity of Chiral compounds.
  • SORareusedto measure the optical activity of a chiral compound and the same is expressed generally in degrees per centimeter per gram per milliliter.
  • DSK uses a Rudolph Polarimeter Autopol IV Automatic with Model: APIV-6W

Karl-Fischer (KF)

  • Karl Fischer titration is a classic titration method that uses coulometric or volumetric titration to determine the trace amounts of water in a compound.
  • DSK uses Lasco KF-Autotitrator.

Lyophilizer

  • Lyophilizer is an instrument which is used to remove the major portion of water and solvents from a compound via controlled freezing and sublimation. It is also used to dry the compounds for storage.
  • DSK uses ZirbusLyophilizer for this purpose.
